The level is done in similar vein to the Donkey Kong Country level Croctopus Chase. Donkey Kong and Diddy Kong will start out underwater, and must avoid some Fangfish and Chomps, until they meet the nautilus enemy named Nemo. The Nemo are shelled creatures that appear to be docile until approached. Then, they emerge from their shell, and chase the Kongs with tentacles extended. The level revolves around swimming away from the Nemo and avoiding enemies as well. Nautilus Chase has several narrow passages which makes it tough to escape.
Also in this level Clambo enemies from DKC are re-introduced as being much larger, but do not have their pearl spitting attack. Instead, the Clambo use their large size to attempt to block Donkey or Diddy from coming through. The clams will simply open and close their mouth in a consistent pattern, and the Kongs must swim by at the right time. In some areas several Clambo can be found. The ending area of the level is a large maze-like area to explore, filled with extra obtainable Bananas and some Chomps Jr. enemies. Swirlwind Storm proceeds this level.

- Nautilus Chase is the first level in Donkey Kong Land whose name is not an alliteration or a rhyme.
