Mario Joe and Luigi Bob are two cousins of Mario and Luigi; described as both hicks and country bumpkins, Mario Joe and Luigi Bob, aside from their clothing, are completely identical to Mario and Luigi.

Near the start of The Super Mario Bros. Super Show! episode "Do You Princess Toadstool Take This Koopa...? / Mario Hillbillies", Mario and Luigi, who were going on a fishing trip, were waiting for both Mario Joe and Luigi Bob to arrive at Mario Brothers Plumbing; eventually, Mario and Luigi, becoming impatient, decided to leave on their fishing trip without Mario Joe and Luigi Bob, who Mario left a note for, explaining where he and Luigi went; it also told Mario Joe and Luigi Bob to make themselves at home in the apartment. Just as Mario and Luigi left Mario Brothers Plumbing through a back entrance, Mario Joe and Luigi Bob arrived in it at the main entrance. Noticing that Mario and Luigi aren't home, Mario Joe and Luigi Bob find Mario's note on the apartment's scarecrow; reading the note aloud, Luigi Bob and Mario Joe, upon reading the part of the note that told them to make themselves at home, instantly let out a yell and ran over to Mario and Luigi's refrigerator, which they began to look through.

Later, Mario Joe and Luigi Bob, having raided Mario and Luigi's refrigerator, began to relax at Mario and Luigi's table, eating large amounts of food. After some small talk involving what pepperoni was and where it came from, Mario Joe and Luigi Bob heard the doorbell of Mario Brothers Plumbing; after some hesitation, Mario Joe and Luigi Bob yelled for whoever was at the door to come in. Approaching the woman who entered the apartment upon hearing them yell, Mario Joe and Luigi Bob learned from the woman that she was looking for a new faucet and asked if Mario Joe or Luigi Bob had one; as Mario Joe tells the visitor that he isn't sure and prepares to also tell her isn't Mario, Luigi Bob stops him and begins to say that he isn't sure if there is a faucet in the plumbing shop at the moment; as the visitor broods over her bad luck, Mario Joe tries to cheer her up, which prompts the visitor to introduce herself as Ellie Mae, who proceeds to ask Mario Joe and Luigi Bob their names. Stuttering, Luigi Bob introduces himself as Luigi, while Mario Joe introduces himself as Mario. Upon learning the two's supposed names, Ellie Mae tells Mario Joe and Luigi Bob that they are just the kind of plumbers she's looking for, country bumpkins, like her.

Afterwards, as Ellie Mae plays with the Ratigator, who Mario Joe and Luigi Bob call Zeke and Luke, Mario Joe and Luigi Bob begin trying to put on tool belts and carry various plumbing tools. As Mario Joe and Luigi Bob struggle, Ellie Mae tells them how kind they were to make a house call for her, but she begins to show doubt in Mario Joe and Luigi Bob's abilities. Reassuring Ellie Mae, Mario Joe and Luigi Bob also tell her that it is an honor to fix her plumbing, which charms Ellie Mae, who decides to have Mario Joe and Luigi Bob stay over at her house for a possum dinner. After Ellie Mae says this, she, Luigi Bob and Mario Joe begin to leave Mario Brothers Plumbing to go to Ellie Mae's house.
