In 2 Player Game, Mario (player one) and Luigi (player two) take turns to complete levels. After one brother completes a level or loses a life, the other brother plays. If one brother interacts with the other brother's icon on the world map, they enter Battle Mode in their Small forms. Whoever wins is immediately given a turn to play in the main game.

Losing Lives and Game Over
Beware! The following are deadly!
Mario will lose one life if he does the following:
If Small Mario takes a hit from an enemy, though if Super Mario is hit by an enemy, he will revert back to Small Mario, If Mario gets damaged while he's powered up, he will only revert to Super Mario [when playing the U.S. version, though in the Japanese version, you would regress all the way back to Small Mario], if Mario gets damaged when wearing the Tanooki, Frog and Hammer suits, the suit will fly off with a "bloop" sound effect and Mario will revert back to Small Mario [When playing the Japanese version, however when playing the U.S. version, the suit disappears in a puff of smoke and Mario just goes back to being Super Mario].
The following will cause instant death:
If Mario falls in a bottomless pit, gets sucked into a suction pit in the water levels, gets sucked into quicksand, or lands in a pool of lava.
If Mario gets sucked into Boss Bass's mouth to become fish food.
If Mario gets squished by auto-scrolling in the auto-scrolling levels.
If the time limit reaches zero [the "TIME-UP!" message is displayed and Mario instantly loses a life]
If Mario loses all of his lives, it's Game Over, and you will go back to the title screen if you choose to end the game, or be sent back to the beginning of the world if you choose to continue, getting a Game Over in the NES version means you have to start all over [Unless if you're playing the SNES and GBA remakes, in which case you can save your progress and start again at the beginning of the world you were in].
And that's how Mario can lose lives.
